The 24-year-old Czech player, who needed wrist surgery a year ago, played a steady and clever game, full of slow slices and angles, to defeat the error-strewn Jabeur, who ended up as the runner-up for the second consecutive year.
Jabeur, who was bidding to become the first Arab player to win a Grand Slam title and the first African woman to lift one of the four major trophies, had 31 unforced errors that hindered her chances.
Vondrousova, who won four matches in the tournament, including Jessica Pegula in the quarter-finals and Elina Svitolina in the semi-finals, celebrated her victory by lifting the Venus Rosewater Dish from Britain's Princess of Wales, appearing in disbelief.
This win makes Vondrousova the fourth Czech-born player to win the title in the professional era after Martina Navratilova, Jana Novotna, and Petra Kvitova.
She is also the sixth successive first-time winner of the women's crown.
